The glycine at codon 276 is replaced by aspartic acid, an amino acid with similar properties. This variant was reported in individual(s) with features consistent with hereditary leiomyomatosis and renal cell cancer (HLRCC) (Ambry internal data). Based on internal structural analysis, this variant is anticipated to result in a significant decrease in structural stability (Ambry internal data; Ajalla Aleixo MA et al. FEBS J. 2019 05;286(10):1925-1940). This variant is considered to be rare based on population cohorts in the Genome Aggregation Database (gnomAD). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis.
