This missense variant replaces histidine with proline at codon 478 of the BRIP1 protein. Computational prediction is inconclusive regarding the impact of this variant on protein structure and function. To our knowledge, functional studies have not been reported for this variant. In a large breast cancer case-control study, this variant was observed in 2/60464 cases and was absent in any controls (PMID: 33471991). This variant has been identified in 1/244232 chromosomes in the general population by the Genome Aggregation Database (gnomAD). The available evidence is insufficient to determine the role of this variant in disease conclusively. Therefore, this variant is classified as a Variant of Uncertain Significance.
This sequence change replaces histidine, which is basic and polar, with proline, which is neutral and non-polar, at codon 478 of the BRIP1 protein (p.His478Pro). This variant is present in population databases (no rsID available, gnomAD 0.0009%). This missense change has been observed in individual(s) with breast cancer (PMID: 34326862).
