The p.G530D variant (also known as c.1589G>A), located in coding exon 15 of the DDX41 gene, results from a G to A substitution at nucleotide position 1589. The glycine at codon 530 is replaced by aspartic acid, an amino acid with similar properties. This variant has been identified in patients with myelodysplastic syndromes and acute myeloid leukemia; the variant was presumed but not confirmed to be of germline origin (Lewinsohn M et al. Blood, 2016 Feb;127:1017-23; Alkhateeb HB et al. Blood Adv, 2022 Jan;6:528-532; Badar T et al. Haematologica, 2023 Nov;108:3033-3043). This amino acid position is highly conserved in available vertebrate species. In addition, this alteration is predicted to be deleterious by in silico analysis. Based on the available evidence, the clinical significance of this variant remains unclear.
This sequence change replaces glycine, which is neutral and non-polar, with aspartic acid, which is acidic and polar, at codon 530 of the DDX41 protein (p.Gly530Asp). This variant is not present in population databases (gnomAD no frequency). This missense change has been observed in individual(s) with acute myeloid leukemia (PMID: 26712909). It has also been observed to segregate with disease in related individuals. ClinVar contains an entry for this variant (Variation ID: 2500222). An algorithm developed to predict the effect of missense changes on protein structure and function (PolyPhen-2) suggests that this variant is likely to be disruptive. In summary, the available evidence is currently insufficient to determine the role of this variant in disease. Therefore, it has been classified as a Variant of Uncertain Significance.
